Products sold as retatrutide in Australia: content against the label

A 2026 letter in Drug and Alcohol Review (PMID 42559975) reports that an accredited laboratory found retatrutide in all three products sold as retatrutide that it tested, at 51.3 percent of the amount stated on the label for one product and at 165 and 190 percent for the other two. Source sentenceResults, Table 1: "One product contained ... (51.3% of labelled content), while the remaining products contained ... (165.0%) and ... (190.0%), respectively."; Methods: NATA accredited laboratory1 Drug Alcohol Rev 2026 Retatrutide is a peptide studied as an agonist at the GLP-1, GIP and glucagon receptors (PubChem CID 171390338, formula C221H342N46O68, about 4731 Da). Source sentenceIntroduction: "Retatrutide is an investigational triple glucagon-like peptide-1, glucose-dependent insulinotropic polypeptide and glucagon receptor agonist currently undergoing clinical evaluation"; PubChem PUG REST: substance records named retatrutide (for example SID 438519203) map to CID 171390338; MolecularFormula C221H342N46O68; MolecularWeight 47311 Drug Alcohol Rev 20262 PubChem CID 171390338

What happened

  • Drug and Alcohol Review published the letter online on 6 Aug 2026, in its September 2026 issue; the authors are at the University of Queensland and PEDTest Australia (PMID 42559975, DOI 10.1111/dar.70231). Source sentencePMC article header: "Drug Alcohol Rev. 2026 Aug 6;45(6):e70231"; PubMed: "Drug Alcohol Rev. 2026 Sep;45(6):e70231"; affiliations School of Public Health, University of Queensland; PEDTest Australia, Perth1 Drug Alcohol Rev 2026
  • The three products were submitted anonymously to PEDTest Australia and analysed by a laboratory accredited by the National Association of Testing Authorities. Source sentenceMethods, paragraph 1: three products sold as retatrutide submitted anonymously to PEDTest Australia; analysed by an independent laboratory accredited by the National Association of Testing Authorities; identity by molecular mass analysis; content quantified per vial; metals by inductively coupled plasma mass spectrometry1 Drug Alcohol Rev 2026
  • Identity was checked by molecular mass, peptide content was quantified per vial, and metals were measured by inductively coupled plasma mass spectrometry.
  • Lead was detected above its quantitation limit; arsenic, cadmium, chromium, nickel and mercury were below their quantitation limits. Source sentenceResults, paragraph 2: "identified no arsenic, cadmium, chromium, nickel or mercury above analytical quantitation limits"; lead "exceeding the practical quantitation limit"1 Drug Alcohol Rev 2026

Why it matters for researchers

  • Identity and content were reported as separate measurements: identity was confirmed in all three samples, while content differed from the label in all three. Source sentenceDiscussion: "all samples contained the expected peptide, but the quantity present varied substantially from the labelled content"1 Drug Alcohol Rev 2026
  • A TGA laboratory test published on 14 Sep 2026 found a different molecule, semaglutide, in a product sold as retatrutide; in this letter the molecule matched and the amount varied. Source sentenceTGA: "the product contained undeclared semaglutide and no retatrutide"3 TGA safety advisory, 14 September 20261 Drug Alcohol Rev 2026

What is not known

  • The letter reports three samples, and the authors state that the findings are not known to reflect the wider market. Source sentenceDiscussion: "Notably, only three samples were analysed, and the findings may not reflect the broader illicit peptide market."1 Drug Alcohol Rev 2026
  • Sterility and endotoxin were not assessed in these samples. Source sentenceDiscussion: "Sterility and endotoxin contamination were not assessed"1 Drug Alcohol Rev 2026
  • The sellers, batches and purchase dates of the three products are not given. Source sentenceMethods and Table 1 list samples 1 to 3 only1 Drug Alcohol Rev 2026
